EZN is an important staining method in the diagnosis of tuberculosis. It is used in the staining of smears prepared to detect the presence of Mycobacteria in clinical samples or culture media. Carbol Fuchsin in EZN dye is a red dye containing Phenol. However, in the EZN staining method, after the Carbol Fuchsin is placed on the smear, it is necessary to heat the smear to liquefy the mycolic acids in order to ensure that the dye is fully infiltrated between the mycolic acids. This is a dangerous operation and has caused laboratory accidents and burns. In order to eliminate this heating process, Kinyoun dye can be used instead of EZN dye. Both phenol and dye concentration are higher in Kinyoun Karbol Fuchsin. In this way, mycobacteria are stained with this dye without the need for heating. In smears stained with EZN stain, mycobacteria are stained red and other microorganisms are stained blue.
