It is used in the staining of smears prepared to detect the presence of mycobacteria in clinical samples or culture media. Fluorochrome staining method using auramine has an important place in the diagnosis of tuberculosis. In microscopic examination, fluorescent stained mycobacteria are detected more easily than bacteria stained with Carbol Fuchsin, making scanning easier and shorter. It should be noted that more false positive results can be obtained in smears examined with fluorescent dye than with Ehrlich Ziehl Neelsen (EZN) or Kinyoun staining methods. Auramine solution is a fluorescent dye solution containing phenol. Mycobacteria give bright yellow green fluorescence in smears stained with auramine dye. Other microorganisms and cell debris are not seen in the examination as they are stained black with potassium permanganate.
